Bacillus megaterium spore germination is influenced by inoculum size.
AIMS The effect of spore density on the germination (time-to-germination, percent germination) of Bacillus megaterium spores on tryptic soy agar was determined using direct microscopic observation. METHODS AND RESULTS Inoculum size varied from approximately 10(3) to 10(8) cfu ml(-1) in a medium where pH=7 and the sodium chloride concentration was 0.5% w/v. Inoculum size was measured by global...
متن کاملSusceptibility of the taro beetle, Papuana uninodis (Coleoptera, Scarabaeidae) to two new Bacillus popilliae isolates from Papuana spp.
Two morphological types of Bacillus popilliae, causal agent of the milky disease, have been isolated from taro beetles (Papuana spp, Coleoptera: Scarabaeidae). B. popilliae from P. woodlarkiana woodlarkiana (Papua New Guinea) was a type A1 with a small sporangium (4.1 x 1.6 microm) and a large spore (2.1 x 1.4 microm) and parasporal body (1.8 x 1.2 microm) that sometimes overlap. B. popilliae f...
متن کاملEffects of sporulation conditions on the germination and germination protein levels of Bacillus subtilis spores.
Bacillus subtilis spores prepared in rich medium germinated faster with nutrient germinants than poor-medium spores as populations in liquid and multiple individual spores on a microscope slide. Poor-medium spores had longer average lag times between mixing of spores with nutrient germinants and initiation of Ca-dipicolinic acid (CaDPA) release. Rich-medium spores made at 37°C germinated slight...
متن کاملA rapid staining technique for the detection of the initiation of germination of bacterial spores.
AIMS We propose to apply the Wirtz-Conklin staining technique to evaluate spore germination. METHODS AND RESULTS Spores at different stages of germination were stained with modified spore stain (Wirtz-Conklin) and evaluated for staining properties. Bacillus spores suspended in deionized water, which does not support germination, stained greenish-blue. Spores suspended in germination enhancers...
متن کاملThe nutritional requirements for germination and outgrowth of spores and vegetative cell growth of some aerobic spore forming bacteria.
During the last 10 years the subject of bacterial spore germination has been intensively investigated. In most of these studies it has been found that bacterial spores are capable of germination under conditions which do not permit the germinated form to survive (Schmidt, 1955; Stedman, 1956). Hills (1949) pointed out that a distinction must be drawn between the requirements for germination and...
